Insulin-like growth factors (IGFs) are similar to insulin by structure and function, but have a much higher growth-promoting activity. They are required for embryonic development, and are produced mainly in the liver in response to a hepatocyte growth hormone. It can be used to substitute insulin for the maintenance of human pluripotent stem cells. IGF-1 LR3 is a recombinant synthetic growth factor of ~9.1 kDa, single polypeptide chain containing 83 amino acid residues. It is an analog of IGF-1 comprised of the complete IGF-1 sequence, with an Arginine substitution for the third position Glutamic acid, and a 13 amino acid length N terminus peptide extension. It has been engineered for higher biological potency in vitro, with an increased half-life.

Bioactivity analysis

The biological activity of CB Recombinant IGF-1 LR3 was tested by its ability to promote cell proliferation, measured using non-radioactive colorimetric assay and monitored by absorbance. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. For this particular case, Core Biogenesis rIGF-1 LR3 outperforms the competitor e.coli derived rIGF-1 standard.

Purity analysis

~ 2 μ g / l a n e o f Recombinant IGF-1 LR3 was resolved in S D S - PA G E reduced ( R ) conditions. The image shows the purity of recombinant I G F - 1 L R 3 with a predicted molecular mass of 9.1 kDa.

Typical Applications

CB recombinant IGF-1 LR3 is generally employed in various stem cell research and cell manufacturing processes. This growth factor delivers superior bioactivity during the maintenance of pluripotent and mesenchymal stem cells, and the differentiation into mesodermal derivatives including chondro, osteo and muscle tissues.
